Cryptotanshinone Induces Apoptosis and Autophagic Cell Death in Prostate Cancer Cells

چکیده
Prostate cancer is one of the most common cancers and one of the leading causes of cancer-related death among men of all races. Complementary and alternative medicine is also used in the treatment of prostate cancer, thus, screening of natural products represents a promising strategy to develop potential anticancer drugs for prostate treatment. Cryptotanshinone, one of the major tanshinones isolated form Danshen, has been reported to inhibit cell proliferation and to induce cell death in various cancer cell lines, including those of colon, cervical, breast, etc. This study aims to explore the cytotoxicity of cryptotanshinone on human prostate cancer cell lines PC3 and DU145 as well as the underlying mechanisms of action. Our results demonstrate the selective cytotoxicity of cryptotanshinone in prostate cancer cells PC3 (IC50 = 15.85 μM) and DU145 (IC50 = 7.59 μM) as well as the underlying mechanisms of action, in particular the activation of both apoptosis and autophagic cell death. These findings suggested that cryptotanshinone is worthy to be further studied for its anticancer potential against prostate cancer.

Epigallocatechin-3-Gallate Induces Apoptosis through Up-regulation of Bax and Down-regulation of Bcl-2 in Prostate Cancer Cell Line
Background and Aims: Epigallocatechin-3-gallate (EGCG) is a polyphenolic compound from green tea, which its anticancer effects on many types of cancers have been confirmed, but the molecular mechanism by which EGCG induces apoptosis remains unknown. The aim of the present study was to investigate anti-proliferative properties and apoptotic signaling pathway of EGCG on PC3 human prostate cancer ...
متن کاملAnticancer Activity of Curcumin-Loaded PLGA Nanoparticles on PC3 Prostate Cancer Cells
Curcumin (Cur) has been found to be very efficacious against many different types of cancercells. However, the major disadvantage associated with the use of Cur is its low systemicbioavailability. Our present work investigated the toxic effect of encapsulation of Cur in PLGA(poly lactic-coglycolic acid) nanospheres (NCur) on PC3 human cancer prostate cell. In thepresent study, we have investiga...

متن کاملCombination of Arsenic trioxide and Everolimus (Rad001) synergistically induces both autophagy and apoptosis in prostate cancer cells
The inhibitor of PI3K-AKT-mTOR pathway, such as Rad001, has not shown therapeutic efficacy as a single agent in prostate cancer. Arsenic trioxide induces the autophagic pathway in prostate cancer cells. We identified Arsenic trioxide can synergize with Rad001 to induce cytotoxicity of prostate cancer cells. Moreover, we identified synergistic induction of autophagy and apoptosis as the underlyi...
متن کاملNovel Poly(Adenosine Diphosphate-Ribose) Polymerase (PARP) Inhibitor, AZD2461, Down-Regulates VEGF and Induces Apoptosis in Prostate Cancer Cells
Background: Prostate cancer (Pca) is a heterogeneous disease, and current treatments are not based on molecular stratification. Poly(adenosine diphosphate [ADP]-ribose) polymerase (PARP) inhibitors have recently been found to be remarkably toxic to cells with defects in homologous recombination, particularly cells with BRCA-mutated backgrounds. Therefore, this preliminary study was designed to ...
